Since the invention of hybridoma cell development in 1975, monoclonal antibodies have proved to be extremely valuable reagents. However, development of hybridomas is labor intensive, time consuming, and expensive. Developed antibodies may fail to meet the original intended applications. GENEWIZ provides custom hybridoma development services to meet your monoclonal antibody needs efficiently, effectively and economically. This service includes -
- Immunization of mice and serum screening
- Hybridoma fusion
- Cloning of hybridoma colonies
- Screening of Hybridoma supernatants
- Expansion of positive clones
Protocol -
Day 0 | | Immunization of 3 mice with antigen |
Day 20 | | ELISA and/or Western blot (if provided) analysis of tail bleeds |
Day 21 | | Hybridoma fusion |
Day 40 | | Identification of positive clones by ELISA |
Day 44 | | Confirmation of positive clones by second round of ELISA and/or Western blot (if provided) |
Day 50 | | Preservation of up to 10 positive clones for each antigen and delivery of 10ml supernatants from these clones for customer testing |
Day 60 | | Expansion of hybridoma clones selected by the customer, up to 5 clones per antigen |
Material Required -
- A completed sample submission form (if the antigen is a recombinant or purified protein, please attach an image of a Coomassie stained SDS-PAGE gel or an equivalent)
- 0.5-1 mg of purified protein (MW. > 30kDa) or conjugated synthetic peptide (>12 amino acids) at a concentration of > 0.2mg/ml. If the antigen is an epitope-tagged recombinant protein (except His-tag), we require submission of 0.2 mg of the same protein with another tag for screening. If the antigen is a conjugated peptide, we require submission of 0.2 mg of the same peptide conjugated to another carrier for screening
- 4 blots for antibody screening at customer's discretion (protocol for blots preparation).
Sample Submission -
Please send proteins by express carriers at conditions similar to the proteins' storage conditions. Please ship proteins that are stored at -80°C on dry ice. Proteins that are stored at -20°C or conjugated peptides in solution can be shipped on blue ice. Most lyophilized antigens can be sent at ambient temperature. Please clearly label the name and the concentration of the proteins.
