Difficult Template Sequencing
DNA Hairpin and RNAi Template Sequencing
RNA Interference (RNAi) refers to the mechanism by which double stranded RNA (dsRNA) specifically suppresses the expression of a gene bearing its complementary sequence. It is an emerging technology with successful applications for quick and definitive studies of gene functions.
RNAi template sequencing, however, can often present challenges due to its double-stranded or "hairpin" structures. The small hairpin RNA (shRNA) templates used in RNAi studies frequently cannot be successfully sequenced, due to early termination or ambiguity. GENEWIZ has developed effective proprietary techniques for sequencing shRNA templates. Our proprietary RNAi Protocol is producing superior results for our customers. For comparison, see the graph below:
GC-Rich DNA Sequencing
GENEWIZ proprietary protocol has been proven successful in sequencing templates with high GC-content and templates with other prohibitive secondary structures.
